Northbound: take highway A1 Rome-Florence and exit at Orte, take road E45 following directions to Perugia and Cesena and exit at Valfabbrica. Exit again at Assisi and follow indications to Assisi

Southbound: take highway A1 Florence-Rome and exit at Valdichiana, take road E45 following directions to Perugia, Cesena and then Terni, exit at Valfabbrica and exit again at Assisi. Follow indications to Assisi.
